CD96, TIGIT, AND PVR:
NEXT-GENERATION CANCER IMMUNOTHERAPY TARGETS
A large number of patients remain disadvantaged by the existing antibody
therapies targeting PD-1, CTLA-4 and PD-L1. The CD96, TIGIT, and PVR/CD155 are
highly expressed in T cells and NK cells, where they regulate these cells'
activation and effector functions. This axis has emerged as a promising
alternative target for immune checkpoint inhibitors. CD96 and TIGIT are in
clinical trials for solid tumors and NSCLC.
PVRIG, CD226, Nectin-1, Nectin-2, Nectin-3, and Nectin-4 are other preclinically
studied ligands and receptors at this axis that modulate T- and NK-cell immune

TIGIT family receptors are a cluster of immunoglobulin superfamily receptors
that interact with the Nectin and nectin-like molecules (Necls) and have
recently emerged as potential targets for cancer immunotherapy. TIGIT, CD226,
CD96, and PVRIG are mainly expressed on activated T cells and NK cells. Their
ligands, PVR, Nectin-1, Nectin-2, Nectin-3, and Nectin-4, are expressed on tumor
cells and antigen-presenting cells (APCs).
As shown in Figure 1, the interactions between the receptors and ligands are
depicted by bilateral arrows, and the arrows are proportional to the affinity of
the interaction.TIGIT has a greater affinity for PVR than CD226 and CD96. The
interaction of Nectin-2 with PVRIG is of higher affinity than with CD226[1,2].
